Sample Submission
Total RNA for microarray experiments must be of high quality:
- 260/280 ratio: 1.8-2.1
- 260/230 ratio: 1.8-2.1
- Bioanalyzer electropherogram showing no degadation
Service Offered
Microarray Experiment
- A Project Request Form must be submitted before
experiments can commence.
- RNA samples are to be submitted in RNase, DNase
free water.
- Typically 1 week turn around time.
- Affymetrix GeneChip experiments require the
submission of one RNA sample for each GeneChip.
- Two aliquots, one labeled "M" for
microarray, a second labeled "B" for Bioanalyzer, of each RNA sample
must be submitted in labeled 1.5 ml microcentrifuge tubes.
- The "M" tube will be used for the
microarray and the second "B" tube will be used for
the Agilent 2100 Bioanalyzer and will contain 5 uL at 200 ng/uL.
3' IVT Expression GeneChips
- Minimum 5 uL of total RNA at 500 ng/uL in RNase, DNase free water.
Exon and GeneArray GeneChips
- Minimum 5 uL of total RNA at 100 ng/uL in RNase, DNase free water.
Cytogenetics Whole-Genome 2.7M Array
- Minimum 6uL of genomic DNA at 33ng/uL (required concentration) in reduced EDTA TE buffer (0.1 mM EDTA, 10mM Tris HCl, pH 8.0).
SNP 5.0/6.0 Mapping GeneChips
- Minimum 12uL of genomic DNA at 50ng/uL (required
concentration) in reduced EDTA TE buffer (0.1 mM EDTA, 10mM Tris HCl, pH 8.0).
500K Mapping GeneChips
- Minimum 6uL (per array) of genomic DNA at 50ng/uL
(required concentration) in reduced EDTA TE buffer (0.1 mM EDTA, 10mM Tris
HCl, pH 8.0).
miRNA GeneChips
- Minimum 8 uL of total RNA at 250 ng/uL in RNase, DNase free water.
Tiling Arrays
- Promoter (single array): 7.5 ug total purified PCR product in a maximum volume of 39.0 uL of RNase, DNase free water.
- Tiling 2.0R (7 arrays sets): 27.0 ug total purified PCR product in a maximum volume of 117.0 uL of RNase, DNase free water.
Agilent 2100 Bioanalyzer Scans
- Used to assess the quality
of total RNA
- Minimum 3 uL of total RNA at 200 ng/uL in RNase, DNase free water.
Spotted cDNA Array Scanning and Image Analysis
- BioRad VersArray ChipReader scanner (5 micron resolution)
- Cy3 and Cy5 filters installed
- Standard microscope slide format
